- Mario - F.L.U.D.D. is now his down B move, while Mario Tornado is his standard aerial-down attack.
- Link - His basic design is based off of Twilight Princess, and thus his weapons and abilities have also slightly changed. His bow has changed into his Hero's Bow and can shoot farther, his boomerang has changed into his Gale Boomerang and can generate whirlwinds, his bombs have changed in appearance, and he can now charge his spin attack while on solid ground.
- Kirby - He greatly resembles the Kirby from the original Super Smash Bros. video game in terms of how he controls.
- Fox McCloud - Not much has changed with this character, though his appearance has been upgraded to suit the one from Command.
- Samus Aran - Is a bit slower, though she can now change into Zero Suit Samus.
- Zelda - Din's Fire is a bit bigger than before, and her overall appearance has changed.
- Bowser - His Fire Breath does not diminish completely, and he's been greatly upgraded.
- In this game, you're allowed to play with four different controllers, including the GameCube controller, the Wii remote and Nunchuck combination, the Wii remote on its side, and the Virtual Console controller.
- Stickers have been added and certainly add a bigger depth to the replay value.
- There are hundreds of trophies, and seemingly a lot more than there were before.
- You're now able to play co-op mode on the home-run contest, Target Smash!!, Multi-Man Brawl, and Events.
- Target Smash!! doesn't feature individual character stages, but instead features 5 levels of target smash stages that can be played with anyone.
- And speaking of the home-run contest -- the stage is now bigger.
- There are new events.
- The Coin Launcher replaces the slot machine from Melee, and is basically a small game within itself.
- The Fighting Alloy Team replaces the Fighting Wire Frames.
- There is now a Stage Builder!
- The adventure mode is much more extensive than before, delivering CGI movies, plenty of bosses (example: Petey Piranha, Rayquaza, etc...), and even a cooperative mode. Now it even has a plot.
- Wi-fi connection has been added!!!
- Final smashes have been added, allowing characters to perform almost instant K.O. attacks.
- You're now able to move, aim and jump while shooting.
- The Footstool Jump allows you to hop off an enemy.
- Gliding will allow particular players to glide in the air.
- There are now three different taunts a player can perform instead of just one.
- You can now place trophies or stickers in certain spots and take a picture of it.
- You can now choose which character you can start out as with Zelda and Pokémon Trainer.
